MAO STAIN

Solutions

• Buffered Formalin
• 900 ml distilled H₂O
• 100 ml 37-40% Formaldehyde
• 6.5 g dibasic Sodium Phosphate
• 4.0 g monobasic Sodium Phosphate
• adjust pH to 7.1
• 1/15M Phosphate Buffer pH 7.6
• 100 ml distilled H₂O
• 0.81 g dibasic Sodium Phosphate
• 0.12 g monobasic Potassium Phosphate
• adjust pH to 7.6 if necessary
• Incubation Medium
• 25 ml 1/15 M Phosphate Buffer pH=7.6
• 75 ml distilled H₂O
• 125 mg tryptamine HCl
• 25 mg nitro-blue tetrazoleum (nitro-BT)

Staining Procedure:

1. Incubate sections in incubation medium at 37° C for 4 hours
2. Fix section in buffered Formalin (5-10 min?)
3. Acetone 15 sec
4. Acetone 30 sec
5. Xylene 1 minute
6. Xylene 2 minutes
7. Coverslip using Dammar Resin
