Golgi Stain

Solutions

• Phosphate Buffer Solution
• 10.934 g dibasic Sodium Phosphate
• 2.76 g monobasic Sodium Phosphate
• 1.0 litre distilled H₂O
• Glutaraldehyde/Formalin/Phosphate(GFP) Solution.
• 350 ml Phosphate Buffer Solution
• 20 ml 25% Glutaraldehyde
• 40 ml 37% Formaldehyde
• 3% Potassium Dichromate Solution
• 3 g Potassium Dichromate
• 100 ml Distilled H₂O
• Store Solution in the dark
• 1% Silver Nitrate Solution
• 1 g Silver Nitrate
• 100 ml Distilled H₂O
• Store Solution in the dark

Procedure

1. Perfuse rat with normal (0.9%) saline.

2. Perfuse rat with GFP Solution

3. Remove brain from cranium and wash briefly in Phosphate Buffer Solution

4. Block brain.

5. Place brain in Potassium Dichromate Solution for 3 days in the dark.

6. Place brain in Silver Nitrate Solution for 3 days in the dark.

Note: When transferring brain from Potassium Dichromate to Silver Nitrate, change Silver Nitrate Solution until no precipitate is seen.
