Golgi Stain
Solutions
- Phosphate Buffer Solution
- 10.934 g dibasic
Sodium Phosphate
- 2.76 g monobasic
Sodium Phosphate
- 1.0 litre distilled H2O
- Glutaraldehyde/Formalin/Phosphate(GFP) Solution.
- 350 ml Phosphate
Buffer Solution
- 20 ml 25% Glutaraldehyde
- 40 ml 37% Formaldehyde
- 3% Potassium Dichromate Solution
- 3 g Potassium
Dichromate
- 100 ml Distilled H2O
- Store Solution
in the dark
- 1% Silver Nitrate Solution
- 1 g Silver Nitrate
- 100 ml Distilled H2O
- Store Solution
in the dark
Procedure
1. Perfuse
rat with normal (0.9%) saline.
2. Perfuse
rat with GFP Solution
3. Remove
brain from cranium and wash briefly in Phosphate Buffer Solution
4. Block
brain.
5. Place
brain in Potassium Dichromate Solution for 3 days in the dark.
6. Place
brain in Silver Nitrate Solution for 3 days in the dark.
Note: When transferring brain from Potassium
Dichromate to Silver Nitrate, change Silver Nitrate Solution until no
precipitate is seen.
